Add Ambion™ RNase H (ribonuclease H) from E. coli overexpressing this gene strains isolated. RNase H specifically degrades RNA in RNA:DNA hybrids to produce products with 3'-hydroxyl and 5'-phosphate terminals. Supplied in a tube containing 1,000 U (10 U/µL). This enzyme does not degrade DNA or unhybridized RNA. RNase H is an integral part of most RNA amplification and NASBA protocols. Can be used to degrade specific RNA when complementary DNA oligonucleotides hybridize (eg, to remove the poly(A) tail from the mRNA hybridized to the oligonucleotide (dT)). Ribonuclease H is rigorously tested for contaminating non-specific endonuclease, exonuclease, RNase and protease activities.
Unit Definition:
One unit of RNase H is measured using 20 pmol of RNaseAlert™ probe coupled to 1,000 pmol of complementary oligonucleotide as substrate. Needle the amount of enzyme required to increase fluorescence by 1.5 RFU per second at 37°C.
